云南昭通天麻多糖含量测定

Determination of Polysaccharides Content in Zhaotong Gastrodia elsta

  • 摘要: 用苯酚-硫酸法对14个昭通天麻样品及3个外购天麻样品进行多糖含量测定。结果显示:在490nm波长处测定吸光度,在浓度0~1120μg/mL之间线性关系良好(r=0.9973);精密度试验,RSD=007%;稳定性试验,RSD=167%;加样回收率1006%,RSD=137%;该方法简单、准确;昭通市彝良县小草坝乡一带所产的天麻样品多糖含量为104%~216%,其多糖含量普遍高于其他地区,可作为判别昭通天麻品质优良的依据之一。

     

    Abstract: The polysaccharides content in 14 samples of Gastrodia elsta produced in Zhaotong and 3 samples of G. elsta bought outside Yunnan Province was determined by means of applying phenolsulfuric acid method. The absorptiophotometric determinations were carried out at 490nm. There was a linear relationship between the photometric value and the concentration within the scale of 0-112.0μg/mL (r=0.9973). The RSD value of precision test was 0.07%, the RSD value of stability test was 167%, and the recovery rate and its RSD value were 1006%and 1.37% respectively. The results of the study indicated that the determination of polysaccharide contents by phenolsulfuric acid method was simple but accurate, the polysaccharides content in the G. elsta produced in Xiaocaoba Township of Yiliang County, Zhaotong Prefecture was 10.4%-21.6%, which was generally higher than those produced from other regions. The polysaccharides content might be adopted as one of the indexes to identify the quality of G. elsta produces.
