Abstract:
BoxBenhnken central composite design and Response Surface Methodology (RSM) were applied to optimize the hydrolytic conditions from homogeneous hydrolysis of βglucosidase by taking enzymatic saccharification of steamexploded corn stover as substrate, and temperature, pH value and βglucosidase dosage as independent variables, while taking the glucose content in the homogeneously hydrolyzed liquor as dependent variable in the experiment. The results showed that the glucose content was significantly affected by temperature and βglucosidase dosages. The optimum conditions for homogeneous hydrolysis of cellobiose enzymatic saccharification were concluded as: 52 ℃ as reaction temperature, pH value 4.0, 1 IU/mL as βglucosidase dosage. Under these conditions, the average glucose content reached 52.81mg/mL in the 3 verification tests.
