秋水仙素处理杉木种子的诱变效应

The Mutagenic Effect of Cunninghamia lanceolata Seed by Colchicine Treatment

  • 摘要: 以杉木成熟种子为试验材料,经25mg/L的赤霉素浸种24h处理后,分别用03%、06%、09%、1.2%的秋水仙素溶液浸泡1、2、3、4d,蒸馏水浸泡种子作为对照,3周后观察统计出苗率、变异率,9个月后调查诱变苗早期长势。结果表明:秋水仙素对种子出苗率、变异率均有显著影响,随着处理浓度和时间的不同呈现出一定规律的变化,09%的秋水仙素浸泡种子4d得到的变异率最大;秋水仙素浓度和处理时间对苗高影响差异显著,同一处理时间随着浓度的增加平均苗高呈现先降低后升高的变化趋势,同一浓度处理则随时间延长苗高表现为先升高后降低的变化趋势;地径大小与苗高规律一致,催芽处理抑制其分枝。

     

    Abstract: The germination rate and mutation rate of the mature seeds of Cunninghamia lanceolata presoaked with 25 mg/L of gibberellin for 24 h were studied by treating with 0.3%, 0.6%, 0.9% and 1.2% colchicine solution for 1d, 2d, 3d and 4d respectively, and the seeds soaked with distilled water were taken as control. The germination rate and mutation rate were observed and counted 3 weeks later, and the mutant seedling growth was observed 9 months later after the treatments. The results indicated that colchicine treatment had a significant influence on seed germination rate and mutation rate, and there was a certain rule of changes in these indexes because of the different colchicine concentration and treatment time. It was showed that the highest mutation rate was resulted from soaking the C.lanceolata seeds for 4 d with 0.9% colchicine. Colchicine concentration and soaking time duration had a significant influence on C.lanceolata seedling height, there was a variation tendency that the average seedling height was firstly reduced and then increased with the increase of colchicine concentration by the same soaking time, while the average seedling height treated by the same colchicine concentration rose up firstly and then dropped afterwards. The change of basal diameter of the C.lanceolata seedling height was consistent with that of the height, and the sprouting treatment inhibited the branching differentiation.
