不同抗生素对紫雨桦离体叶片愈伤组织诱导的影响

Effects of Different Antibiotics on Induction of Callus from the Leaves of Betula pendula ‘Purple Rain’

  • 摘要: 以紫雨桦无菌叶片为试材,研究头孢霉素、头孢曲松钠、羧苄青霉素和特美汀4种不同浓度的抑菌抗生素对紫雨桦离体叶片愈伤组织诱导的影响,分析4种抑菌抗生素对农杆菌EHA105(pCHS)的抑菌效果。结果表明,抗生素是头孢霉素、头孢曲松钠、特美汀比羧苄青霉素抑菌效果好,其中头孢霉素和头孢曲松钠的适宜浓度范围为200~400mg/L、特美汀为400~800mg/L。30~40mg/L的卡那霉素、15~20mg/L的G418或潮霉素还可用于紫雨桦为受体的遗传转化中筛选转化体。

     

    Abstract: The effects of four kinds of antibacterial antibiotics, (Cefotaxime, Ceftriaxone, Carbenicillin and Timentin) on induction of callus were studied using the leaves of Betula pendula ‘Purple Rain’ as explants, and the effects of the antibacterial antibiotics on inhibiting Agrobacterium EHA105(pCHS) were also analyzed. The results indicated that the best antibacterial antibiotics were Cephalosporins, Ceftriaxone and Timentin in Agrobacteriummediated transformation. The appropriate concentration of Cefotaxime or Ceftriaxone was 200-400mg/L, and Timentin was 400-800mg/L. The results also showed that 30-40mg/L kanamycin, 15-20mg/L G418 or Hygromycin were the ideal pressure to screen transformant for Betula pendula ‘Purple Rain’.
