Abstract:
The effects of four kinds of antibacterial antibiotics, (Cefotaxime, Ceftriaxone, Carbenicillin and Timentin) on induction of callus were studied using the leaves of Betula pendula ‘Purple Rain’ as explants, and the effects of the antibacterial antibiotics on inhibiting Agrobacterium EHA105(pCHS) were also analyzed. The results indicated that the best antibacterial antibiotics were Cephalosporins, Ceftriaxone and Timentin in Agrobacteriummediated transformation. The appropriate concentration of Cefotaxime or Ceftriaxone was 200-400mg/L, and Timentin was 400-800mg/L. The results also showed that 30-40mg/L kanamycin, 15-20mg/L G418 or Hygromycin were the ideal pressure to screen transformant for Betula pendula ‘Purple Rain’.
