不同产地金线莲组培快繁试验

Study on Tissue Culture and Rapid Propagation of Anoectochilus Roxburghii in Different Origins

  • 摘要: 以不同产地金线莲幼嫩茎段为外植体诱导丛生芽,通过不同培养基配方的筛选,建立不同产地金线莲组培快繁体系。结果表明,不同产地金线莲最佳初代培养基分别是,福建永安产地为MS+20mg/L 6-BA+02mg/L NAA+10mg/L KT,台湾南投产地为MS+30mg/L 6-BA+02mg/L NAA+10mg/L KT,广西玉林产地为MS+40mg/L 6-BA+02mg/L NAA+10mg/L KT;最佳继代培养基分别是,福建永安产地为MS+20mg/L 6-BA +02mg/L NAA,台湾南投产地为MS+30mg/L 6-BA +03mg/L NAA,广西玉林产地为MS+20mg/L 6-BA+02mg/L NAA;在壮苗培养阶段,使用外源添加物蛋白胨后,不同产地金线莲外植体苗的生长状况在不同程度上得到了改善,最佳生根培养基分别是,福建永安产地为1/2MS+15mg/L NAA +05mg/L IBA+20mg/L ABT1,台湾南投产地为1/2MS+05mg/L NAA+05mg/L IBA,广西玉林产地为1/2MS +05mg/L NAA+10mg/L IBA+10mg/L ABT1,同时添加15g/L Ac;炼苗移栽基质V(蛭石)∶V(泥炭土)=1∶1,移栽生根率均达90%以上。

     

    Abstract: To obtain the system of tissue culture and rapid propagation of Anoectochilus roxburghii from different origins, the tender stem segments of A. roxburghii from different origins were taken as explant, and screened optimum culture medium formula to induce shoot clusters. The results showed that: the optimum initial mediums of A. roxburghii in Fujian, Taiwan and Guangxi was MS+20 mg/L 6-BA+02 mg/L NAA+10 mg/L KT, MS+30 mg/L 6-BA+02 mg/L NAA+10 mg/L KT, and MS+40 mg/L 6-BA+02 mg/L NAA+10 mg/L KT, respectively In addition, the optimum subculture medium of A roxburghii in Fujian, Taiwan and Guangxi was MS+20 mg/L 6-BA+02 mg/L NAA for, MS+30 mg/L 6-BA+03 mg/L NAA for, and MS+20 mg/L 6-BA+02 mg/L NAA, respectively In the stage of seedling cultivation, all plants from different producing areas could be improved growth in different degrees after applying exogenous peptone Additional, the optimum rooting medium A roxburghii in Fujian, Taiwan and Guangxi was 1/2MS+15 mg/L IBA+05 mg/L NAA+20 mg/L ABT, 1/2MS+05 mg/L IBA+05 mg/L NAA, and 1/2MS+05 mg/L IBA+ 10 mg/L NAA+10 mg/L ABT1+ 15 g/L Ac Meanwhile, plantlets from test tube grew well on the growth medium of vermiculitethe and peat soil mixed at the rate of 1∶1, the rooting rate of the A roxburghii reached 90%
