橡胶树乳管表达多酚氧化酶基因HbPPO1的克隆与分析

Molecular Cloning and Analysis of a Polyphenol Oxidase Gene (HbPPO1) from the Hevea brasiliensis Laticifer

  • 摘要: 根据EST序列设计引物,应用RACE和RT-PCR技术从橡胶树的胶乳(乳管细胞的胞质成分)中分离到1条长2102bp的cDNA,该序列包含1个1803bp的开放读码框(ORF),81bp的5’UTR和218bp的3’UTR;ORF预测编码600个氨基酸,其理论分子量为6788kDa,等电点为856,质体定位;蛋白含有多酚氧化酶特有的Tyrosinase、PPO1_DWL和PPO1_KFDV结构域;蛋白与木薯、蓖麻、麻疯树和胡杨中同源蛋白的相似性分别为852%、820%、798%和782%,将基因命名为HbPPO1。结果表明,在所分析的胶乳、树皮和叶片组织中,基因在胶乳中的表达丰度最高,且其表达水平受乙烯利显著抑制。

     

    Abstract: Based on the EST sequence, a 2 102 bp cDNA was isolated from the rubber tree (Hevea brasiliensis) latex (representing the laticicfer cytoplasm) with specific primers via RACE and RT-PCR. The sequence designated as HbPPO1 contains a 1 803 bp long open reading frame (ORF), 81 bp 5′ UTR and 218 bp 3′ UTR. Sequence analysis indicated that HbPPO1 putatively encodes 600 amino acids with a theoretical molecular weight (Mw) of 67.88 kDa and isolectric point (pI) of 8.56, which was predicted to be localized to the plastid. The protein contains the conserved Tyrosinase, PPO1_DWL and PPO1_KFDV domains specific to polyphenol oxidases, and shares the similarity of 85.2%, 82.0%, 79.8% and 78.2% with its homologs in Manihot esculenta, Ricinus communis, Jatropha curcas and Populus euphratica. Expression analysis showed that, among the examined tissues such as laticifer, bark and leaf, HbPPO1 expressed relatively more in the laticifer and the expression level was significantly inhibited by ethylene.
