不同葡萄霜霉病抗性品种叶内生细菌群落分析
Analysis of Leaf Endophytic Bacterial Communities in Grape Varieties with Different Levels of Downy Mildew Resistance

摘要: 为探究葡萄霜霉病抗性差异与叶片内生细菌群落的关联性,采用16S rDNA高通量测序技术,比较6种葡萄健康叶片内生细菌群落的多样性、组成和结构差异。结果表明:6个品种在物种丰富度和群落结构上差异显著,其中抗病品种“阳光玫瑰”的特有OTUs数量、物种丰富度及群落多样性均显著高于感病品种“蓝宝石”。抗病品种分别富集鞘氨醇单胞菌属、黄杆菌属、泛菌属及芽孢杆菌属等潜在抗病相关菌属。LEfSe分析表明,差异物种在两个抗病品种中分别特异性富集。其中,肠杆菌目、盐单胞菌科、Candidatus Portiera属及海洋螺菌目显著富集于“阳光玫瑰”;而“黑皇”中特异性富集的物种较少,仅检出立克次体目。葡萄叶片内生细菌群落特定菌属的富集特征与霜霉病抗性密切相关,不同抗病品种可能通过特异性菌群增强抗性。Abstract: To investigate the association between differences in resistance to grape downy mildew and endophytic bacterial communities in leaves, 16S rDNA high-throughput sequencing technology was employed to compare the diversity, composition, and structural differences of endophytic bacterial communities in healthy leaves of 6 grape varieties. The results showed significant differences in species richness and community structure among the 6 varieties. Among them, the number of unique OTUs, species richness, and community diversity of the disease-resistant variety 'Shine Muscat' were significantly higher than those of the susceptible variety 'Sweet Sapphire'. Disease-resistant varieties enriched potential disease resistance-related genera such as Sphingomonas, Flavobacterium, Pantoea, and Bacillus. LEfSe analysis indicated that differential species were specifically enriched in two disease-resistant varieties. Among them, o__Enterobacterales, f__Halomonadaceae, Candidatus Portiera, and o__Oceanospirillales were significantly enriched in 'Shine Muscat'; while fewer specifically enriched species were detected in 'Black Emperor', with only o__Rickettsiales identified. This study suggests that the enrichment characteristics of specific genera in the endophytic bacterial community of grape leaves are closely related to downy mildew resistance, and different disease-resistant varieties may enhance resistance through specific microbial communities.
